Establishment of the Department of Bone Barrow Transplantation & Cancer Immunotherapy; Israel’s National Bone Marrow Transplantation Center and the Cell Therapy & Transplantation Research Laboratory focusing on combining basic science and experimental medicine towards development of new therapeutic strategies for regulation of immunohematopoietic system aiming for: [1] Up-regulation of the immune system for immunotherapy of cancer by cytokines, adoptive allogeneic cell therapy focusing on activated and targeted donor lymphocytes; [2] Down-regulation of the immune system focusing on new approaches for induction of self tolerance and tolerance to alloantigens for prevention of rejection and graft-versus-host disease (GVHD) and for induction of transplantation tolerance for organ allografts; [3] Development of new approaches for safer and more effective stem cell transplantation, focusing on non- myeloablative conditioning for the treatment of cancer, genetic and autoimmune diseases; [4] new approaches for cancer immunotherapy focusing on cell therapy, vaccine therapy and using targeted anti-cancer agents (small molecules; targeted intentionally mismatched killer cells and cancer-seeking viruses).
More recently, the research laboratory was engaged in exploiting the use of bone marrow stem cells in the context of stem cells plasticity and tissue repair. We have introduced the use of mixtures of bone marrow cells and demineralized bone matrix, when indicated together with new reverse thermal gelation polymers for induction of new bone and cartilage formation for orthopedic, neurosurgical and dental transplantation indications. In parallel, we have introduced new methods for stem cell transplantation directly into the intra-osseous space and induction of donor derived bone marrow microenvironment. This approach will provide an option for safer and more effective transplantation of stem cells for elderly and high risk patients, especially those with diseases associated with abnormal bone marrow microenvironment, such as myelofibrosis. Preliminary experimental data suggest that these new approaches may also facilitate induction of mixed chimerism and transplantation tolerance.
In an attempt to discover more effective and safer approaches for control of self-reactivity for treatment of autoimmune diseases and alloreactivity for control of rejection and GVHD, new families of molecules developed by chemists working in our laboratory are currently under investigation, aiming for induction of antigen-specific unresponsiveness by small molecules. In parallel, we are now synthesizing new small molecules for control of cancer cells, focusing on tyrosine kinase and other signal transduction signal inhibitors on the one hand, and agents like ceramide and sphingolipid analogues for induction of apoptosis, on the other (patents pending and new applications in progress).
Starting in 2007:
Continuation of applied research in cancer treatment and regenerative medicine and clinical application of innovative medical procedures involving the use of cell therapy for the treatment of malignant and non-malignant disorders, including regenerative medicine with stem cells products at the ICTC in Tel Aviv.
